Decontamination and disposal of used media.

1.0 Objective
To lay down the procedure to describe the decontamination and disposal of used media and used items in modren microbiology ltd.

2.0 Scope
This procedure is applicable for all used media, expired media, and used items for decontamination processes and disposal in the Microbiology Department of modrenmicrobiology ltd.

3.0 Responsibility
3.1 Microbiologist / Trained Personnel:
Responsible for following the procedure as per this SOP.

4.0 Accountability
4.1 Head/Designee – Microbiology:
Accountable for ensuring adherence to this SOP.

5.0 Procedure
5.1 After the completion of incubation and observations, carry the used media and items to the decontamination autoclave room (G107/A).

5.2 Wear a nose mask and hand gloves while handling used media.

5.3 Place used liquid media bottles with growth and oncology drugs into the autoclave basket for decontamination.

5.4 Remove solidified agar from bottles using a spatula and place it in a biohazard bag.

5.5 Place petri plates, Bioball vials, cryovials, biological indicator ampoules, and other miscellaneous items in a biohazard bag.

5.6 Tie the biohazard bags with a cable tie and affix a chemical indicator to the bag.

5.7 Run the autoclave according to SOP No.: SOP/MMB/015. Parameters:

Temperature: 121°C – 124°C
Pressure: 1.2 kg/cm² (15 psi)
Duration: 60 minutes
5.8 Load the autoclave as per Load Pattern.

5.9 After autoclaving, check the chemical indicator for color change (off-white to black).

If no change is observed, restart the cycle with a new chemical indicator.
5.10 Transfer autoclaved liquid media into biohazard bags and add 100 mL disinfectant solution (daily prepared) to each bag. Mix thoroughly, then seal the bags again.

5.11 Allow the media to cool to room temperature.

5.12 Place decontaminated items into a yellow-colored poly bag. Label the bag with the date and store it in the “Used Microbiology Media – Biohazard” drum.

5.13 Decontaminated items should be transferred through the Dynamic Pass Box to the Biohazard Waste Storage Room.

5.14 Record decontamination details in the logbook and send the items to the Safety Department for safe disposal.

5.15 Enter operation details into the Autoclave Logbook.

5.16 In case of spills, follow cleanup procedures.

5.17 Wash glassware as per glassware cleaning SOP.

5.18 Disposable garments, lint-free cloths, gloves, and other items used for handling oncology drugs must be packed in double disposable bags and handed over to the Safety Department for incineration.

5.19 General waste shall be sent to authorized third-party facilities for incineration through the Safety Department.
