Microbiological Analysis of Compressed Air/Nitrogen Gas

1. OBJECTIVE:
1.1. To lay down the Procedure for Microbiological Analysis of Compressed Air/Nitrogen Gas in
Modern microbiology Ltd.

2. SCOPE:
2.1. This procedure is applicable to microbiology department in Modern microbiology Ltd.

3. RESPONSIBILITY:

3.1. Microbiology personnel shall be responsible for this SOP.

3.2. Engineering personnel shall be ensure the sampling activity.

4. ACCONTABILITY:

4.1. Microbiology HOD/designee

5. PROCEDURE:
5.1. Sampling procedure:
5.1.1. Before performing the activity AR Number shall be raised in sample inward and record the reconciliation
details in media usage log book..
5.1.2. Use 90 mm SCDA Plates for collection of compressed air /nitrogen gas samples.
5.1.3. Check the calibration status of air sampler, media expiry date and accessories expiry date for before
proceedings the sampling.
5.1.4. Sanitize the external surface of media plates, Accessories and Air sampler with 70%IPA.
5.1.5. Sanitize SS bin with 70%IPA place the media plates and Accessories in to SS bin
5.1.6. carry the air sampler and SS bin to sampling location by following respective entry exit procedure
5.1.7. Mark the medial plates with AR Number, date of sampling and name of location.
5.1.8. Engineering personal shall ensure the flow rate of compressed air/nitrogen gas.
5.1.9. Microbiology person fit the y shape cone tube end to compressed air/ nitrogen gas outlet.
5.1.10. Place the media plate in air sampler and close the air sampler with Y shape cone head then start the air
sampler to suck the air up to 1000/m3
5.1.11. Ensure the flow rate 1ltr/ min is within the limit or not.
5.1.12. After collection of 1000 ltr of air, remove the media plate carefully and keep the media plates in SS
bin.
5.1.13. Bring the SS Bin to microbiology lab and incubate the plates as per incubation period.
5.2. Incubation &observation:
5.2.1. Incubate the all plates along with one negative control plate in 30-35 c for 48 hrs. After completion of
incubation record the transfer details in respective format and shift the plates to 20-25c for 72 hrs.
5.2.2. After the completion of incubation conditions take the plates from incubator and record the incubation
completion details in respective format.
5.2.3. Count the colonies by using colony counter and record the results in respective format.
5.3. Acceptance criteria:
5.3.1. Total microbial count:100CFU/ m3
5.4. Frequency:
5.4.1. Once in a year with in±15 days.
